Melior's Approach to in vivo Models.

Melior's core philosophy is that comprehensively evaluating a compound in vivo requires a complete understanding of a compound's pharmacokinetic and pharmacodynamic properties. Without adequate consideration of these properties prior to designing the experimental protocol, a good deal of the drug candidate's potential may be overlooked.

Therefore, following determination of a maximum tolerated dose, all compounds are routinely tested for dose-response relationships across a 3-point curve, using the route of administration appropriate for the particular compound's in vivo properties. Melior routinely evaluates an array of pharmacokinetic parameters for test compounds such as Tmax, Cmax, t1/2 (alpha, beta), and VD, prior to entering the compound into the theraTRACE® platform. Working interactively with the collaboration partner, Melior selects a dosing regimen suited for each particular compound and each in vivo model.

This careful planning around optimal route of administration, dose and formulation, coupled with continuous monitoring of plasma levels during the course of the study, significantly enhances the power of the theraTRACE® platform.
